本研究根据genbank中查出的禽流感病毒h _ 5亚型，禽流感病毒h _ 9亚型的ha片段基因组序列，利用dnasis软件分别对aiv的h _ 5 、 h _ 9亚型ha基因区域进行同源性比较，设计筛选出两对联合pcr反应的特异性引物，其中一对是h _ 9亚型通用引物，扩增出ha片段695bp ，另一对引物为h _ 5亚型的通用引物，扩增出的ha区域部分目的片段约为448bp 。
